چکیده

Stem cell regulatory mechanisms are difficult to study because self-renewal and production of differentiated progeny, which are both strictly controlled, occur simultaneously in these cells. To focus on the self-renewal mechanism alone, we investigated the behavior of germinal stem cells (GSCs) in progeny-deficient testes with defective GSC differentiation. In these testes, we found that the proliferation of undifferentiated spermatogonia, some of which are GSCs, was accelerated by high concentrations of glial cell line-derived neurotrophic factor (GDNF). Furthermore, we found that follicle-stimulating hormone (FSH) stimulation via homeostatic control was one of the major regulators of GDNF concentration. These results suggest that in mammalian testes, GSC proliferation and population size are regulated homeostatically by the GDNF/FSH pathway.

Expression of Spermatogonial and Pluripotency Markers in Spermatogonial Stem Cells after Treatment with Different Culture Factors

Background: As condition and component of culture determine fate map of spermatogonial stem cells (SSCs), the aim of this study was to evaluate of growth factors GDNF, LIF and RA on proliferation and differentiation of SSC. Materials and Methods: SSCs were cultured in two groups: The first group GDNF and LIF and the second group RA. The number of clumps and colony formation was monitored dur...

Glial cell line-derived neurotropic factor stimulates sertoli cell proliferation in the early postnatal period of rat testis development.

Glial cell line-derived neurotropic factor (GDNF) was first characterized by its actions on central nervous system neurons. GDNF messenger RNA was expressed in many peripheral tissues in addition to brains. We demonstrated that GDNF had a marked stimulatory effect on the proliferation of Sertoli cells during the early postnatal period of rat testis development. The action of GDNF was examined i...

The Effects of Plla Nanofiber Scaffold on Proliferation of Frozen-Thawed Neonate Mouse Spermatogonial Stem Cells

Purpose: To investigate of the effects of a poly L-lactic acid (PLLA) nanofiber scaffold on proliferation of frozen-thawed neonate mouse spermatogonial stem cells.Materials and Methods: Spermatogonial cells were isolated from neonatal 3-6-day-old NMRI mice testes by two steps enzymatic digestion and differential plating. The isolated spermatogonial cells were divided into four culture groups: 1...

Effect of Lithium Chloride on Proliferation and Bone Differentiation of Rat Marrow-Derived Mesenchymal Stem Cells in Culture

Objective(s) It is believed that the mesenchymal stem cell (MSC) differentiation and proliferation are the results of activation of wnt signaling pathway. On the other hand, lithium chloride is reported to be able to activate this pathway. The objective of this study was to investigate the effect of lithium on in vitro proliferation and bone differentiation of marrow-derived MSC. Materials and ...

The Luteinizing Hormone-Testosterone Pathway Regulates Mouse Spermatogonial Stem Cell Self-Renewal by Suppressing WNT5A Expression in Sertoli Cells

Spermatogenesis originates from self-renewal of spermatogonial stem cells (SSCs). Previous studies have reported conflicting roles of gonadotropic pituitary hormones in SSC self-renewal. Here, we explored the role of hormonal regulation of SSCs using Fshb and Lhcgr knockout (KO) mice. Although follicle-stimulating hormone (FSH) is thought to promote self-renewal by glial cell line-derived neuro...
